요약The intervertebral disc (IVD), also known as the spinal disc, undergoes accelerated degeneration compared to other connective tissues, leading to intervertebral disc degeneration. Addressing this challenge requires effective treatments capable of reversing fibrocartilage damage caused by factors such as age and injury. Tissue-resident stem cells hold promise for IVD treatment due to their self-renewal and differentiation properties. In this dissertation, I will discuss our efforts to identify novel regulators of spinal skeletal stem cell (SSC) differentiation and their role in regenerating IVD tissue in response to injury. Additionally, I will outline a protocol we developed for isolating and functionally assessing human SSCs from various cell and tissue sources. Our basic and preclinical findings lay the foundation for innovative stem cell-based therapies targeting IVD degeneration and other related disorders.
