요약The overall objectives of this dissertation were to study how nutrients, specifically vitamin D, and social grouping contributes to the immune function, health, growth and development of dairy cattle. Chapter 1 reviews the literature on the mastitis and heifer development and roles of vitamin D and calf housing on dairy calf development and immunity. Chapters 2 and 3 pertain to the effects of intramammary vitamin D treatments. Chapter 2 reports the results of testing the effect of dose and source of intramammary vitamin D treatments on gene expression and serum calcium. The work from Chapter 2 shows that 500 ug intramammary calcidiol (25-hydroxyvitamin D3) influenced milk cell gene expression without affecting serum Ca; whereas 50 ug calcitriol (1,25-dihydroxyvitamin D3) influenced milk cell gene expression but also increased serum Ca.Chapter 3 reports the effectiveness of intramammary vitamin D as a treatment for mastitis in dairy cattle. Calcitriol treatment administered at the onset of mastitis symptoms decreased the percentages of CD11b+CD14-(neutrophils) and concentrations of 8-iso-15R isoprostane in milk of infected quarters compared with placebo. Calcitriol did not affect bacteria or somatic cell 13counts in milk. The intramammary calcitriol treatment was not able to remove the S. uberis infection. Chapters 4 and 5 focus on heifer calf growth and immunity. Chapter 4 presents the effects of dietary calcidiol (1.7 ug/kg BW) on growth, health, and performance of dairy heifers until 3 months of age. Supplementing dietary calcidiol increased average body weight by 4 kg at weaning compared to control heifers possibly due to increased milk intake. Calcidiol decreased the concentration of monocytes in blood. Chapter 5 reports the effect of dairy calf housing on calf performance and immune competence. Housing treatments had no effect on calf body weights or average daily gain at weaning. Granulocyte percentage was decreased in GH calves indicating lower presence of infection. The concentration of CD T cells in blood was influenced by the interaction of social housing by age. Cell-mediated ex vivo recall responses to C. Albicans were influenced by social housing. Likewise, antibody-mediated responses to Keyhole limpet hemocyanin were influenced by the interaction of social hosing and age. Altogether, vitamin D nutrition and social housing are factors in immune development of dairy calves.
